    Technical note:The persistence of microbial-specific DNA sequences through gastric digestion in lambs and its potential use as microbial markers

    Two groups of 5 lambs were euthanized at the weaning (T45) and fattening stages (T90) to evaluate the use of microbial ribosomal DNA (rDNA) sequences as potential microbial markers in relation to purine bases (PB) as a conventional marker. Both microbial markers originated similar microbial N concentrations (mg/g of DM), although T45 showed decreased values compared with the T90 group when either PB or rDNA were considered (P = 0.02). The survival of microbial rDNA was determined in 3 digestive sites (omasum, abomasum, and duodenum), but no substantial differences were observed, indicating that rDNA maintains the molecular stability along the sampling sites analyzed. Contrarily PB concentration increased successively along the digestive tract (P < 0.05), likely as a consequence of the endogenous PB secretion. Undegraded milk PB may also explain the overestimation of the microbial N concentration (2.8 times greater) using PB than rDNA sequences. Abomasum was the sampling site where the best agreement between PB and rDNA estimations was observed. Protozoal N concentration was irrelevant in T45 animals, although substantial in T90 lambs (18% of microbial N). In conclusion, bacterial 16S and protozoal 18S rDNA sequences may persist through the gastric digestive tract and their utilization as a highly specific microbial marker should not be neglected.This study was supported by a FPU grant from the Education and Science Spanish Ministry (project: AGL 2004-02910/GAN) and by a University of Zaragoza project (UZ2008-BIO-04)

    Modeling experimental parameters for the fabrication of multifunctional surfaces composed of electrospun pcl/zno-nps nanofibers

    In this work, a one-step electrospinning technique has been implemented for the design and development of functional surfaces with a desired morphology in terms of wettability and corrosion resistance by using polycaprolactone (PCL) and zinc oxide nanoparticles (ZnO NPs). The surface morphology has been characterized by confocal microscopy, scanning electron microscopy (SEM), atomic force microscopy (AFM) and water contact angle (WCA), whereas the corrosion resistance has been evaluated by Tafel polarization curves. Strict control over the input operational parameters (applied voltage, feeding rate, distance tip to collector), PCL solution concentration and amount of ZnO NPs have been analyzed in depth by showing their key role in the final surface properties. With this goal in mind, a design of experiment (DoE) has been performed in order to evaluate the optimal coating morphology in terms of fiber diameter, surface roughness (Ra), water contact angle (WCA) and corrosion rate. It has been demonstrated that the solution concentration has a significant effect on the resultant electrospun structure obtained on the collector with the formation of beaded fibers with a higher WCA value in comparison with uniform bead-free fibers (dry polymer deposition or fiber-merging aspect). In addition, the presence of ZnO NPs distributed within the electrospun fibers also plays a key role in corrosion resistance, although it also leads to a decrease in the WCA. Finally, this is the first time that an exhaustive analysis by using DoE has been evaluated for PCL/ZnO electrospun fibers with the aim to optimize the surface morphology with the better performance in terms of corrosion resistance and wettability.Project RTI2018-096262-B-C41–MAITAI, funded by MCIN/AEI/10.13039/501100011033 and, as appropriate, by ERDF 'A way of making Europe'.     Se ha realizado un estudio mineralógico semicuantitativo, de 12 polvos de talco de uso tópico lubricante; elegidos como muestra representativa de este tipo de productos en farmacias y droguerias españolas. El contenido de talco oscila entre 70-95%, con un contenido medio de 85,2 %. El talco medio contiene diversos impurificantes en las siguientes cantidades: 2,7 % de clorita, 3,3 % de calcita, 1,5 % de dolomita, 1 % de anfíboles, 0,72 % de cuarzo y 3,4 % de caolinita.It has been effectued a semicuantitative mineralogical analysis of 12 dusting powder talc to topic use. These samples were selected asmore representative of these products procedents from several spanish chemists. The content of tale is compressed· beneath 70 and 95 %. , with an average content of 85,2% . The medium tale content several impurities with following quantities: 2,7 % of chlorite, 3,3 % of calcite, 1,5 % of dolomite, 1 % of amphiboles, 0,72 % of quartz and 3,4 % of kaolinite

    COVID-19 Severity and Survival over Time in Patients with Hematologic Malignancies: A Population-Based Registry Study

    Mortality rates for COVID-19 have declined over time in the general population, but data in patients with hematologic malignancies are contradictory. We identified independent prognostic factors for COVID-19 severity and survival in unvaccinated patients with hematologic malignancies, compared mortality rates over time and versus non-cancer inpatients, and investigated post COVID-19 condition. Data were analyzed from 1166 consecutive, eligible patients with hematologic malignancies from the population-based HEMATO-MADRID registry, Spain, with COVID-19 prior to vaccination roll-out, stratified into early (February–June 2020; n = 769 (66%)) and later (July 2020–February 2021; n = 397 (34%)) cohorts. Propensity-score matched non-cancer patients were identified from the SEMI-COVID registry. A lower proportion of patients were hospitalized in the later waves (54.2%) compared to the earlier (88.6%), OR 0.15, 95%CI 0.11–0.20. The proportion of hospitalized patients admitted to the ICU was higher in the later cohort (103/215, 47.9%) compared with the early cohort (170/681, 25.0%, 2.77; 2.01–3.82). The reduced 30-day mortality between early and later cohorts of non-cancer inpatients (29.6% vs. 12.6%, OR 0.34; 0.22–0.53) was not paralleled in inpatients with hematologic malignancies (32.3% vs. 34.8%, OR 1.12; 0.81–1.5). Among evaluable patients, 27.3% had post COVID-19 condition. Buenas prácticas para la integración académica y personal del estudiante Erasmus en el Grado en Español: Lengua y Literatura(Fase I), desarrollado en la Facultad de Filología de la Universidad Complutense de Madrid durante el curso académico 2018-2019.     Association of Candidate Gene Polymorphisms With Chronic Kidney Disease: Results of a Case-Control Analysis in the Nefrona Cohort

    Chronic kidney disease (CKD) is a major risk factor for end-stage renal disease, cardiovascular disease and premature death. Despite classical clinical risk factors for CKD and some genetic risk factors have been identified, the residual risk observed in prediction models is still high. Therefore, new risk factors need to be identified in order to better predict the risk of CKD in the population. Here, we analyzed the genetic association of 79 SNPs of proteins associated with mineral metabolism disturbances with CKD in a cohort that includes 2, 445 CKD cases and 559 controls. Genotyping was performed with matrix assisted laser desorption ionizationtime of flight mass spectrometry. We used logistic regression models considering different genetic inheritance models to assess the association of the SNPs with the prevalence of CKD, adjusting for known risk factors. Eight SNPs (rs1126616, rs35068180, rs2238135, rs1800247, rs385564, rs4236, rs2248359, and rs1564858) were associated with CKD even after adjusting by sex, age and race. A model containing five of these SNPs (rs1126616, rs35068180, rs1800247, rs4236, and rs2248359), diabetes and hypertension showed better performance than models considering only clinical risk factors, significantly increasing the area under the curve of the model without polymorphisms. Furthermore, one of the SNPs (the rs2248359) showed an interaction with hypertension, being the risk genotype affecting only hypertensive patients. We conclude that 5 SNPs related to proteins implicated in mineral metabolism disturbances (Osteopontin, osteocalcin, matrix gla protein, matrix metalloprotease 3 and 24 hydroxylase) are associated to an increased risk of suffering CKD

    Predictive Power of the "Trigger Tool" for the detection of adverse events in general surgery: a multicenter observational validation study

    Background In spite of the global implementation of standardized surgical safety checklists and evidence-based practices, general surgery remains associated with a high residual risk of preventable perioperative complications and adverse events. This study was designed to validate the hypothesis that a new “Trigger Tool” represents a sensitive predictor of adverse events in general surgery. Methods An observational multicenter validation study was performed among 31 hospitals in Spain. The previously described “Trigger Tool” based on 40 specific triggers was applied to validate the predictive power of predicting adverse events in the perioperative care of surgical patients. A prediction model was used by means of a binary logistic regression analysis. Results The prevalence of adverse events among a total of 1,132 surgical cases included in this study was 31.53%. The “Trigger Tool” had a sensitivity and specificity of 86.27% and 79.55% respectively for predicting these adverse events. A total of 12 selected triggers of overall 40 triggers were identified for optimizing the predictive power of the “Trigger Tool”. Conclusions The “Trigger Tool” has a high predictive capacity for predicting adverse events in surgical procedures. We recommend a revision of the original 40 triggers to 12 selected triggers to optimize the predictive power of this tool, which will have to be validated in future studies
